We describe a 3D culture model of the human breast epithelium that is suitable to study hormone action.
The process of mammary epithelial morphogenesis is influenced by hormones. The study of hormone action on the breast epithelium using 2D cultures is limited to cell proliferation and gene expression endpoints. However, in the organism, mammary morphogenesis occurs in a 3D environment. 3D culture systems help bridge the gap between monolayer cell culture (2D) and the complexity of the organism. Herein, we describe a 3D culture model of the human breast epithelium that is suitable to study hormone action. It uses the commercially available hormone-responsive human breast epithelial cell line, T47D, and rat tail collagen type 1 as a matrix. This 3D culture model responds to the main mammotropic hormones: estradiol, progestins and prolactin. The influence of these hormones on epithelial morphogenesis can be observed after 1- or 2-week treatment according to the endpoint. The 3D cultures can be harvested for analysis of epithelial morphogenesis, cell proliferation and gene expression.
In tegenstelling tot standaard 2D culturen 3D celkweek surrogaat apparaten kan de studie van epitheelcel gedrag in een fysiologisch relevante context, iemand als een weefsel. 3D culturen van de borstklier geholpen verhelderen vele aspecten van ontwikkeling van de borstklier en neoplasie. De meeste van de 3D cultuurmodellen momenteel beschikbaar zijn ongeschikt hormoonactie bestuderen omdat het menselijke epitheliale cellijnen gebruikt voor de taak missen hormoonreceptor expressie 6,7,9.
Hierin beschrijven we een 3D cultuurmodel van de menselijke borst epitheel die geschikt is om hormooninwerking 12 bestuderen. Dit model maakt gebruik van de commercieel verkrijgbare hormoon reagerende menselijke borst epitheel cellijn, T47D 3,11,13, die oorspronkelijk waren afgeleid van een pleurale effusie verkregen uit een 54 jaar oude vrouwelijke patiënt met een infiltrerend ductaal carcinoom van de borst. We maken gebruik van rattenstaart collageen type 1 als een matrix. Deze 3D culture model is geschikt voor de studie van de werking van de drie mammotropic hormonen (estradiol, promegeston (een analoog van progesteron) en prolactine) menselijke borst epitheelcellen. Hormoon-geïnduceerde epitheliale morfologie kan kwantitatief worden geëvalueerd na verloop van tijd door morfometrische analyse 12.
Een geschikte zaaidichtheid kunnen deze 3D kweken gedurende 2 weken worden bewaard. Tegen die tijd, de ontwikkeling van structuren is voldoende voor een robuuste kwantitatieve beoordeling van hormoonwerking op epitheliale morfologie. Gels kunnen ook worden geoogst eerdere tijdstippen voor celproliferatie en genexpressie analyses. Daarnaast heeft dit model geschikt om de effecten van een sequentiële hormonale behandeling getest; bijvoorbeeld na behandeling met estradiol gedurende de eerste week en vervanging door andere hormonen / combinatie van hormonen in de volgende week. Het effect van oestrogene stoffen en anti-oestrogenen, zoals ICI 182.780, kan worden stustierf het gebruik van deze 3D-cultuur model 12.
Here, we describe a hormone-sensitive 3D culture model to test the action of hormones on breast epithelium.
