We beschrijven hier een protocol voor de zuivering van zeer actieve Hsp104, een hexamere AAA + eiwit van gist, die koppels ATP hydrolyse aan eiwit desaggregatie. Deze regeling maakt gebruik van een His6-gelabeld bouwen voor affiniteitszuivering uit<em> E. coli</em> Gevolgd door anion-uitwisseling chromatografie, His6-tag verwijderen met TEV protease, en de grootte-uitsluiting chromatografie.
