This manuscript presents a unique in vitro model of immunopurified human villous cytotrophoblast cells cultured under hypoxia/reoxygenation. This model is suitable to study the protective effects of promising treatments, such as melatonin, on pregnancy complications associated with increased oxidative stress and altered placental function.
This protocol describes how villous cytotrophoblast cells are isolated from placentas at term by successive enzymatic digestions, followed by density centrifugation, media gradient isolation and immunomagnetic purification. As observed in vivo, mononucleated villous cytotrophoblast cells in primary culture differentiate into multinucleated syncytiotrophoblast cells after 72 hr. Compared to normoxia (8% O2), villous cytotrophoblast cells that undergo hypoxia/reoxygenation (0.5% / 8% O2) undergo increased oxidative stress and intrinsic apoptosis, similar to that observed in vivo in pregnancy complications such as preeclampsia, preterm birth, and intrauterine growth restriction. In this context, primary villous trophoblasts cultured under hypoxia/reoxygenation conditions represent a unique experimental system to better understand the mechanisms and signalling pathways that are altered in human placenta and facilitate the search for effective drugs that protect against certain pregnancy disorders. Human villous trophoblasts produce melatonin and express its synthesizing enzymes and receptors. Melatonin has been suggested as a treatment for preeclampsia and intrauterine growth restriction because of its protective antioxidant effects. In the primary villous cytotrophoblast cell model described in this paper, melatonin has no effect on trophoblast cells in normoxic state but restores the redox balance of syncytiotrophoblast cells disrupted by hypoxia/reoxygenation. Thus, human villous trophoblast cells in primary culture are an excellent approach to study the mechanisms behind the protective effects of melatonin on placental function during hypoxia/reoxygenation.
Hela graviditet, placenta cytotrofoblastceller, som är mononukleära stamceller, snabbt föröka sig och differentiera till antingen villösa eller extravillous cytotrofoblastceller. Extravillous cytotrofoblaster invadera och omforma spiral artärerna i livmoderväggen. Villösa cytotrofoblaster, å andra sidan, fortsätter att föröka sig, differentiera och säkring för att bilda flerkärniga syncytiotrofoblast (syncytium) 1. Upprätthållandet av villi trofoblasten homeostas är viktigt för fostrets välbefinnande och hälsosam graviditet. I själva verket, villösa trofoblaster tillåter moder till foster utbyte av syre och näringsämnen, och producerar viktiga hormoner för graviditet. Dessutom är syncytiotrofoblast den enda celltyp i direkt kontakt med det maternella blodcirkulationen och ger en väsentlig fysisk och immunologisk barriär. Därför måste syncytiotrofoblast genomgå apoptos och ersättning för homeostatisk underhåll och AVOid placenta patologier 2-5.
Tekniken har utvecklats av Kliman et al. 6 i 1986 för att isolera primära villösa cytotrofoblaster från humana moderkakor orsakade en revolution i placenta forskning genom att studera de molekylära mekanismer som är involverade i villi trofoblasten differentiering. Denna klassiska teknik, baserad på sekventiella enzymatiska spjälkningar med trypsin och DNas, följt av isolering i densitetscentrifugering media (kolloidalt kiseldioxidpartiklar belagda med polyvinylpyrrolidon, eller Percoll) erkänns nu som den gyllene standarden för att isolera villösa cytotrofoblastceller. Tekniken kan optimeras genom magnetisk immunorening, ett förfarande som skiljer villösa cytotrofoblaster från icke-trofoblastiska celler baserat på det differentiella uttrycket av specifika antigener på ytan av dessa celler. Vi valde den HLA-antigen ABC (HLA-ABC) på grund av frånvaron av dess uttryck på trofoblastisk cell membrane 7,8.
Moderkakan är ett organ som genomgår dramatiska variationer i syrenivåer under graviditeten. I den första trimestern, är syreförhållandet fysiologiskt mycket låg (2% O 2) men ökar till milda nivåer av syresättning (8% O 2) i den andra och tredje trimestern. Et al. Tuuli 9 beskrev att reproduktionen av trofoblasten miljön inne i placenta villi in vitro är en utmaning och variationer i syresättningsnivåer kan även leda till fenotypiska förändringar. Det är därför föreslog att anta 8% syre som normoxi att efterlikna syretrycket finns i placenta villi under tredje trimestern av graviditeten 8,9. Chen m.fl.. 10 studerats ingående flera variabler relaterade till syrespänning i trofoblaster cellkultur och visat på vikten av att bestämma syrehalten i en pericellulär miljö. Halterna av syre i villi tenderar att ökapå grund av vaskulogenes. Blodflödet i moderkakan villi ökar ständigt och nivån av väteperoxid (en riklig reaktiva syreradikaler) är en viktig signal som styr vaskulogenes 11,12. I graviditetskomplikationer, en brist på vaskulogenes genererar hypoxi, och ännu viktigare, intermittenta variationer av syresättning (kallas hypoxi / syresättningen). Dessa förhållanden leder till en onormal ökning av oxidativ stress, vilket äventyrar placenta och fetal livskraft 13,14. De förändringar som trofoblastceller genomgår in vivo under episoder av hypoxi / ny syresättning kan efterliknas in vitro enligt följande: villösa cytotrofoblaster hålls under normoxiska förhållanden (8% O 2) tills de differentieras till syncytiotrofoblast. De utsätts sedan för hypoxiska betingelser (0,5% O 2) under 4 timmar, följt av ytterligare 18 h av normoxi (ny syresättning). Med hjälp av denna hypoxi / ny syresättning tillvägagångssätt, trofoblaster exHibit avreglerad redox status och ökade nivåer av inneboende apoptos 8, vilket har observerats i vissa komplikationer. Därför är detta en användbar in vitro-modell för att utvärdera nya förebyggande och terapeutiska metoder för att bekämpa komplikationer i samband med placenta hypoxi / syresättningen.
Placentaceller producerar melatonin, som har flera viktiga funktioner, såsom en förmåga att undvika oxidativ stress och placenta dysfunktion 15. Hos däggdjur är fosterutveckling direkt beroende av korrekt placenta funktion.
