Abstract

中药材质量控制是中药材研发的重要组成部分。面对中草药现代化、国际化的挑战,迫切需要建立全面有效的中药材质量鉴定程序,迫切需要高效、准确、环保的新型分析和检测技术。

多重光散射是一种尖端的分析方法,可以在不改变样品的性质或状态或使用有机试剂的情况下准确、快速地评估中草药的质量。Indigo Naturalis 被认为是治疗小儿体温过高、牛皮癣、白血病和溃疡性结肠炎的良好药物。在这项研究中,使用多重光散射仪精确记录了在水中添加 Indigo Naturalis 粉末的过程。

通过仪器的定性和定量测量,可以准确捕捉天然靛蓝粉体入水的整体轨迹和下沉行为,建立以样品透射和后向散射光谱图为定性指标,稳定性指标为定量指标的天然靛蓝品质快速评价方法。基于多重光散射的分析技术,为天然靛蓝的质量评价提供了一种快速、准确、绿色、环保的方法,支撑着高品质靛蓝天然靛蓝的开发和转化。

Introduction

在中医药中,在疾病治疗过程中,药物的临床有效性和治疗过程的安全性直接受到中草药质量的影响。利用尖端的识别技术,可以评估中草药的疗效并保证使用者的安全。中草药水质检测法是指将中药浸入水或溶剂中,然后通过观察颜色、大小和形状的变化,快速准确地确定药物的真实性1.

本来是鉴定中药的好选择。然而,传统水质检验方法的缺点是,由于肉眼观察的主观性,鉴别中药真伪的准确性和灵敏度较低2。水测试方法中使用的关键药材之一是 Indigo Naturalis,被认为是治疗小儿体温过高、牛皮癣、白血病和溃疡性结肠炎的有效药物3.正宗的Indigo Naturalis漂浮在水面上,摇晃后水不会变成深蓝色。然而,假的 Indigo Naturalis 有下沉的颗粒,摇晃后水会变成深蓝色4.其原理是由于疏水且易于漂浮的靛蓝、靛蓝和优质天然靛蓝的其他有机成分。相反,由于有机物含量低、石灰含量大、质地较重,一些掺有假靛蓝天然靛蓝的颗粒会很快下沉5.然而,这种方法只是简单的定性鉴定,限制了中草药真伪的快速鉴定,无法揭示靛蓝在水中的变化。

多重光散射技术是一种基于激光穿过样品来测量多角度光信号扫描的技术。入射光在穿透样品或遇到颗粒时会被散射。如果散射光穿透样品,则形成透射光信号;如果样品浓度高,光会被颗粒反射,形成反向散射光信号。光强的变化反映了液体制备中颗粒浓度和粒径的变化6.多重光散射仪可通过多重光散射技术快速准确地分析悬浮液、乳液、泡沫液的乳化、絮凝、析出、破裂等现象,并定量分析上述现象的发生率等特征。

多重光散射技术在颗粒稳定性监测7、红酒澄清8 和牛奶发酵质量控制9 方面显示出显著优势。使用这项技术,Indigo Naturalis 的传统水质测试方法可能是直观、定量和科学的。因此,本研究基于多重光散射技术原理,以样品的Turbiscan稳定性指数(TSI)为质量控制指标,建立了一种对Indigo Naturalis品质的快速评价方法(图1)。

Protocol

1.测试样品的制备 设置四个不同批次的 Indigo Naturalis 草本粉进行测试。依次将每个样品通过第七筛和第九筛,并在第七和第九筛5之间收集样品。注意:第七个筛子的平均孔径尺寸为 125 μm ± 5.8 μm。第九筛的平均孔径尺寸为75μm±4.1μm。 在称量纸上准确称取 0.2 g 样品(Indigo Naturalis 粉末)并将其放在一边。 2. 样品添加</…
