Een protocol om trichothecenen (mycotoxinen van zorg voor de gezondheid van de mens) met behulp van een nieuw ontwikkelde screening methode gebaseerd op een competitief immunochemische methode en een laatste elektrochemische detectie te detecteren is aangetoond.
Immunoassays zijn een goed alternatief voor de duurdere en tijdrovende kwantitatieve HPLC of GC 1, 2 methoden voor het screenen detectie van gevaarlijke mycotoxinen in levensmiddelen. In dit protocol laten we zien hoe te fabriceren en te ondervragen een elektrochemische competitieve enzyme linked immunomagnetische assay gebaseerd op het gebruik van magnetische beads als vaste ondersteuning voor de immunochemische keten 3 en gezeefdrukt elektroden sensing platform.
Onze methode heeft tot doel vast te stellen het totale bedrag van de HT-2 en T-2-toxine, mycotoxinen die behoren tot de trichothecenen familie en van groot belang voor de gezondheid van de mens 4. Het gebruik van een antilichaam kloon met een kruis reactiviteit van 100% naar HT-2 en T-2 maakt het mogelijk om gelijktijdig zowel de toxines te detecteren met een vergelijkbare gevoeligheid 5.
De eerste stap van onze test is de coating stap waar we HT2-KLH conjugaat toxine immobiliseren op het oppervlak van de magnetische kralen. Na een blokkerende stap die nodig is om niet-specifieke absorptie te voorkomen, de toevoeging van een monoklonaal antilichaam kan de concurrentie tussen de geïmmobiliseerde HT-2 en gratis HT-2 of T-2 aanwezig is in het monster of opgelost in een standaard oplossing.
Aan het einde van de wedstrijd stap, het bedrag van monoklonale antilichaam gekoppeld aan het geïmmobiliseerde HT-2 zullen worden omgekeerd evenredig aan de hoeveelheid gif in de monsteroplossing.
Een secundaire antilichaam gelabeld met alkalische fosfatase (AP) wordt gebruikt om de binding tussen de specifieke antilichaam en het geïmmobiliseerde HT-2 te onthullen. De laatste meting stap wordt uitgevoerd door het droppen van een hoeveelheid van magnetische kraal ophanging, wat overeenkomt met een specifiek monster / standaard oplossing, op het oppervlak van een gezeefdrukte werkende elektrode, magnetische korrels worden geïmmobiliseerd en geconcentreerd door middel van een magneet precies onder de gezeefdrukt elektrode. Na twee minuten incubatie tussen magnetische kralen en een substraat voor AP, is de enzymatische product gedetecteerd door Differential Pulse voltammetrie (DPV) met behulp van een draagbaar instrument (PalmSens) ook in staat om automatisch acht metingen te starten binnen een interval van enkele seconden.
Het gebruik van antilichamen als biomoleculaire erkenning probe heeft gezien een wijdverbreide gebruik voor het waarnemen van technologieën; immunochemische detectie methoden, zoals ELISA's en MEIAs, zijn tegenwoordig een van de meest gebruikte platforms en toegepast in veel laboratoria 7.
Hoewel deze benaderingen uitzonderlijke gevoeligheid en specificiteit, een primaire doelstelling van vele onderzoeksgroepen te bereiken in de afgelopen jaren is het verbeteren en optimalise…
