Протокол для обнаружения трихотецены (микотоксины заботы о здоровье человека), используя недавно разработанный метод скрининга на основе конкурентных иммунохимического метода и окончательного выявления электрохимический продемонстрировано.
Иммунологического являются реальной альтернативой более дорогим и трудоемким количественных ВЭЖХ или GC 1, 2 методы скрининга обнаружения опасных микотоксинов в пищевых товаров. В этом протоколе мы покажем, как изготовить и допросить электрохимической конкурентной Фермент связан иммуномагнитный анализ, основанный на использовании магнитных шариков в виде твердых поддержку иммунохимических цепочке 3 и трафаретной печатью электродов в качестве чувствительных платформы.
Наш метод имеет целью определить общую сумму НТ-2 и Т-2 токсины, микотоксины, принадлежащих трихотецены семьи и большой опасностью для здоровья человека 4. Использование антител клон с перекрестной реактивности на 100% по отношению к НТ-2 и Т-2 позволяет одновременно обнаруживать как токсины с аналогичными чувствительности 5.
Первым шагом нашего анализа является покрытие шаг, где мы обездвижить HT2-KLH сопряженных токсина на поверхности магнитных бус. После блокировки шаг, необходимо, чтобы избежать неспецифического поглощения, добавление моноклональных антител позволяет конкуренция между иммобилизованным НТ-2 и свободной HT-2 или Т-2 присутствует в образце или растворенные в стандартное решение.
В конце конкурса шаг, количество моноклональных антител связан с иммобилизованным НТ-2 будет обратно пропорциональна количеству токсина в растворе образца.
Вторичных антител, меченных щелочной фосфатазы (AP) используется, чтобы выявить связь между специфических антител и иммобилизованных НТ-2.
