Med hjälp av mänskliga primära prostataepitelceller, rapporterar vi en ny biomarker-fri metod för funktionell karakterisering av stamceller med en sfäroid-baserade etikett-retention assay. Ett steg-för-steg-protokoll beskrivs för BrdU, CFSE eller långt röd 2D-cellmärkning; tredimensionell sfäroid formation; etikett-behålla Stem-liknande cell identifiering av immunocytochemistry; och isolering av FACS.
