Here we describe a procedure allowing a detailed analysis of the phosphorylation-dependent activation of the IRF3 transcription factor. This is achieved through the combination of a high resolution SDS-PAGE and a native-PAGE coupled to immunoblots using multiple phosphospecific antibodies.
The IRF3 transcription factor is critical for the first line of defense against pathogens mainly through interferon β and antiviral gene expression. A detailed analysis of IRF3 activation is essential to understand how pathogens induce or evade the innate antiviral response. Distinct activated forms of IRF3 can be distinguished based on their phosphorylation and monomer vs dimer status. In vivo discrimination between the different activated species of IRF3 can be achieved through the separation of IRF3 phosphorylated forms based on their mobility shifts on SDS-PAGE. Additionally, the levels of IRF3 monomer and dimer can be monitored using non-denaturing electrophoresis. Here, we detail a procedure to reach the highest resolution to gain the most information regarding IRF3 activation status. This is achieved through the combination of a high resolution SDS-PAGE and a native-PAGE coupled to immunoblots using multiple total and phosphospecific antibodies. This experimental strategy constitutes an affordable and sensitive approach to acquire all the necessary information for a complete analysis of the phosphorylation-mediated activation of IRF3.
De alomtegenwoordig en constitutief uitgedrukt transcriptiefactor Interferon (IFN) Regulatory Factor 3 (IRF3) is van cruciaal belang voor de eerste verdedigingslinie tegen pathogenen voornamelijk door de inductie van IFNβ, maar ook door de inductie van de chemokine (CC-motief) ligand 5 (CCL5 ) en verscheidene antivirale eiwitten zoals IFN geïnduceerde eiwitten met tetratricopeptide herhaalt IFIT1 / 2/3 1-3. IRF3 activering werd gemeld na infectie met verschillende virussen, of blootstelling aan polyinosinic-polycytidylic acid (poly I: C) of lipopolysaccharide (LPS) 4. C-terminale fosforylering van IRF3 impliceert niet minder dan 7 verschillende phosphoacceptor locaties georganiseerd in twee clusters, Ser385 / Ser386 en Ser396 / Ser398 / Ser402 / Thr404 / Ser405, die elk bijdragen aan IRF3 activering door middel van dimerisatie, nucleaire accumulatie, associatie met het CREB -bindende eiwit (CBP) / p300 coactivatoren, DNA-bindende gevoelige respons element (ISRE) consensus sequenties en transactivatie van target genen 9,10,17-19 IFN. Fosforylering van Thr390 is ook gedacht om bij te dragen aan-virus-geïnduceerde IRF3 activering 20. Massaspectrometrie analyses van IRF3 hebben aangetoond dat Ser386, Thr390, Ser396 en Ser402 resten direct fosforylatieed door de remmer van KB-kinase ε (IKKε) /-TANK bindend kinase 1 (TBK1) kinasen 9,10. Fosforylering van het C-eindstandige resten is ook vereist voor beëindiging van IRF3 activering door middel polyubiquitinatie en proteasoom-afhankelijke afbraak 10. Deze werkwijze is ook afhankelijk van de fosforylatie van Ser339, die nodig is voor het aantrekken van de propyl isomerase Pin1 10,11. IRF3 species met tenminste fosfo-Ser339 / 386/396 residu's worden beschouwd gehyperfosforyleerd vormen. De exacte volgorde en de functie van elke site blijft een onderwerp van discussie 10,21. Het is nu duidelijk dat geactiveerd IRF3 vormt geen homogene staat, maar dat de verschillende geactiveerde soorten vertonen duidelijke fosforylering of dimerisatie kenmerken bestaan 10,22.Om een volledig begrip van IRF3 activering mogelijk in antwoord op specifieke pathogenen, is het dus noodzakelijk om characterize welke de geactiveerde species worden opgewekt. Inductie van IRF3 target genen, IFNB1 es IFIT1, heeft zich bewezen als een betrouwbare uitlezing voor IRF3 activering mogelijk maken. De controle expressie van deze genen geen onderscheid maakt tussen verschillende toestanden van activering IRF3. Een uitgebreide analyse van IRF3 activering staten in een bepaalde instelling is gebaseerd op de gedetailleerde karakterisering van de fosforylering en dimerisatie-status 10. Gefosforyleerde (formulier I), gehypofosforyleerde (vorm II) en hypergefosforyleerde (vormen III en IV) IRF3 vormen 6,18,23 succes kan worden opgelost door een verminderde mobiliteit in hoge-resolutie SDS-PAGE analyse. Monomeer en dimeer IRF3 soorten kunnen efficiënt worden geïdentificeerd door native-PAGE analyse. Deze benaderingen zijn sterk verbeterd bij gebruik in combinatie met fosfospecifiek antilichamen gericht tegen verschillende IRF3 phosphoacceptor sites.
Standaardprotocollen maken een slechte resolutie vaneiwitten die efficiënte scheiding van verschillende IRF3 gefosforyleerde vormen niet toestaat. Hier hebben we in vivo onderscheid tussen de verschillende geactiveerd in detail beschrijven een procedure om de hoogste resolutie bereiken de inductie van afzonderlijke-virus geactiveerd IRF3 soort, volgens SDS-PAGE gekoppeld met natieve PAGE in combinatie met immunoblot gebruikt totaal fosfospecifiek antilichamen te controleren. vormen van IRF3 wordt uitgevoerd op basis van hun mobiliteit verschuivingen waargenomen op SDS-PAGE. Bovendien kan IRF3 monomeer en dimeer worden onderscheiden door niet-denaturerende elektroforese. Het protocol beschrijven we hier bestaat uit een combinatie van hoge-resolutie SDS-PAGE en natieve PAGE gekoppeld aan het gebruik van verschillende fosfospecifiek antilichamen tegen het monomeer / dimeer en phosphoforms I-IV van IRF3 onderscheiden. Geschikte detectie van deze IRF3 soort is essentieel om volledig te karakteriseren IRF3 activering in een specifieke instelling.
