Vår studie fokuserar på effekterna av leptin signalering i halspulsådern kroppen (CB) på hypoxisk andningshjälp svar (HVR). Vi utförde “förlust av funktion” experiment mäta effekten av leptin på HVR efter CB denervering och “vinst av funktion” experiment mäta HVR efter överuttryck av leptin receptorn i CB.
Ett adipocyte-producerat hormon leptin är en potent respiratorisk stimulerande, som kan spela en viktig roll för att försvara andningsfunktionen hos fetma. Den halspulsådern organ (CB), ett nyckelorgan i perifer hypoxisk känslighet, uttrycka den långa funktionella isoform av Leptin receptor (LepRb) men rollen av leptin signalering i kontroll av andning har inte helt klarlagd. Vi undersökte den hypoxiska andningshjälp responsen (HVR) (1) hos C57BL/6j möss före och efter infusion med leptin vid baseline och efter CB denervation; (2) i leprb-bristfällig feta db/DB möss vid baseline och efter Leprb överuttryck i CBs. I C57BL/6J möss ökade leptin HVR och effekterna av leptin på HVR avskaffades av CB denervation. I db/DB- möss har leprb -uttrycket i CB förstärkt HVR. Därför, vi drar slutsatsen att leptin agerar i CB att öka Svaren till hypoxi.
En fettceller produceras hormon leptin verkar i hypotalamus att undertrycka födointag och öka ämnesomsättningen. Studier utförda i vårt laboratorium1,2 och av andra utredare3,4 visade att leptin ökar hyperkapnisk andningshjälp respons (HVR) för att förhindra obesitas hypoventilation i Leptin brist på fetma. Emellertid, en majoritet av överviktiga individer har höga halter av leptin i plasma och uppvisar resistens mot de metabola och respiratoriska effekterna av hormonet5,6,7,8. Resistens mot leptin är multifaktoriell, men begränsad permeabilitet av blod-hjärnbarriären (BBB) till leptin spelar en viktig roll. Vi föreslår att leptin verkar under BBB i ett nyckelorgan av perifer hypoxisk känslighet, halspulsådern organ (CB), att försvara andning hos överviktiga individer. CBs uttrycker den långa funktionella isoformen av Leptin receptor, leprb, men rollen av CB i respiratoriska effekter av leptin har inte varit tillräckligt klarlagd9,10.
Målet med vår metod var att undersöka effekten av leptin signalering i CB på HVR. Vår motivering var att utföra (a) förlust av funktions experiment infusion leptin hos möss med intakta karotikroppar och denervated halspulsådern organ följt av HVR mätningar; b) vinst av funktions experiment i db/DB- möss som saknar leprb, där vi mätte HVR vid baseline och efter uttryck av LEPRb uteslutande i CB. Fördelen med vår teknik var att vi utförde alla våra experiment i ohämmade tre möss under sömnen och vakenhet. Tidigare utredare utförde antingen sina experiment under anestesi9 eller inte mäta effekterna av leptin under sömn10. Dessutom är vår studie den första att använda en unik vinst av funktionssätt med selektiv LepRb uttryck i CB som beskrivs ovan.
I det breda sammanhanget, vår metod kan generaliseras till andra receptorer uttryckta i CB och deras roll i hypoxisk känslighet. Utredare kan ingjuta en ligand till en receptor av intresse och mäta HVR vid baseline och efter CB denervation. Som en kompletterande metod kan en receptor av intresse överuttryckas i CB och HVR mätningar kan utföras före och efter överuttryck med hjälp av vår teknik som beskrivs i detta manuskript.
Huvudfokus i vår studie var att undersöka luftvägs effekter av leptin signalering i CB. Flera protokoll har utvecklats för att bedöma rollen av leptin på ett mekanistiskt sätt. För det första analyserades det specifika bidraget från CB till HVR genom noggrann kvantifiering av HVR under den första 2 min av hypoxisk exponering.
