Nervbanor är topografiskt organiserade i funktionella fack med specifika molekylära profiler. Här ger vi de praktiska och tekniska åtgärder för att avslöja den globala hjärnan topografi med hjälp av en mångsidig wholemount immunhistokemisk färgning strategi. Vi demonstrerar användbarheten av metoden med användning av väl förstått cellarkitekturen och kretsarna i lillhjärnan.
Den upprepade och väl förstod cellulär arkitektur lillhjärnan gör det till en idealisk modell för att utforska hjärnan topografi. Bakom den relativt enhetliga cellarkitekturen är en komplex uppsättning parasagittal domäner gen-och proteinuttryck. Den molekylära uppdelning av lillhjärnan speglas av anatomiska och funktionella organisation afferenta fibrer. För att till fullo uppskatta komplexiteten i lillhjärnan organisation vi förfinat tidigare en wholemount färgning metod för hög genomströmning analys av mönstring defekter i musen lillhjärnan. Detta protokoll beskriver i detalj de reagens, verktyg och praktiska åtgärder som är användbara för att framgångsrikt avslöja mönster proteinuttryck i den vuxna möss lillhjärnan med wholemount immunfärgning. De steg som lyfts fram här visar nyttan av denna metod att använda uttrycket av zebrinII / aldolaseC som ett exempel på hur den fina topografi hjärnan kan avslöjas i sinnativa tredimensionella konformation. Också beskrivet är anpassningar till protokollet att möjliggöra visualisering av proteinuttryck i afferenta prognoser och stora cerebella för jämförande studier av molekylära topografi. För att illustrera dessa program är data från afferenta färgning av råttcerebellum ingår.
Vi har beskrivit de tekniska uppgifter som krävs för en lyckad wholemount färgning med hjälp av en mångsidig immunhistokemisk metod för avslöjande protein uttryck i utvecklande och vuxna hjärnan.
