We developed a gene-chimeric preparation of ventral hippocampal – accumbens circuit in vitro that allows direct live imaging to analyze presynaptic mechanisms of nicotinic acetylcholine receptors (nAChRs) mediated synaptic transmission. This preparation also provides an informative approach to study the pre- and post-synaptic mechanisms of synaptic plasticity.
Sustained enhancement of axonal signaling and increased neurotransmitter release by the activation of pre-synaptic nicotinic acetylcholine receptors (nAChRs) is an important mechanism for neuromodulation by acetylcholine (ACh). The difficulty with access to probing the signaling mechanisms within intact axons and at nerve terminals both in vitro and in vivo has limited progress in the study of the pre-synaptic components of synaptic plasticity. Here we introduce a gene-chimeric preparation of ventral hippocampal (vHipp)–accumbens (nAcc) circuit in vitro that allows direct live imaging to analyze both the pre- and post-synaptic components of transmission while selectively varying the genetic profile of the pre- vs post-synaptic neurons. We demonstrate that projections from vHipp microslices, as pre-synaptic axonal input, form multiple, reliable glutamatergic synapses with post-synaptic targets, the dispersed neurons from nAcc. The pre-synaptic localization of various subtypes of nAChRs are detected and the pre-synaptic nicotinic signaling mediated synaptic transmission are monitored by concurrent electrophysiological recording and live cell imaging. This preparation also provides an informative approach to study the pre- and post-synaptic mechanisms of glutamatergic synaptic plasticity in vitro.
Cholinerge modulatie circuit exciteerbaarheid bij aan fundamentele aspecten van cognitie en veranderde cholinerge modulatie is een kenmerk van neurodegeneratieve en neuropsychiatrische stoornissen, waaronder de ziekte van Alzheimer, ziekte van Parkinson, schizofrenie en verslaving 1-4. Een gevestigde mechanisme cholinerge vergemakkelijking van synaptische transmissie in het centrale zenuwstelsel is via directe activatie van nAChRs gelokaliseerd op pre-synaptische plaatsen. Activatie van deze pre-synaptische receptoren leidt tot intracellulaire Ca2 + ([Ca2 +] i) in presynaptische terminals verhoogd – zowel direct, door de relatief hoge geleiding door calcium bepaalde nAChR subtypes en indirect via intracellulaire signalering cascades 5, en zodoende neurotransmitter release. In feite is de activatie van presynaptische nAChRs in verband met veranderingen in afgifte van diverse neurotransmitters zoals glutamaat, GABA, ACh, eennd dopamine 6-10. Hoewel deze werkwijze is onderzocht Indirect door elektrofysiologische werkwijzen op diverse synapsen, optische reporters van [Ca2 +] i en synaptische vesicles recycling mogelijk directer en tijdelijk nauwkeurige meting van presynaptische verschijnselen.
Pre-synaptische lokalisatie van nAChRs is overtuigend aangetoond met directe immuno-goud etikettering van nAChRs bij het elektron microscopische (EM) niveau 11,12. Verscheidene andere technieken zijn ook gebruikt voor de nAChR lokalisatie indirect pakken, onder detectielocaties van nAChRs subunit- fluorescerend eiwit chimaera in gekweekte neuronen 13,14, elektrofysiologische registratie van nAChR stromen in synaptische terminals 15,16, bewaken nicotine geïnduceerde veranderingen in [Ca 2+] i in synaptische zenuwuiteinden door live cell imaging 17, en indirecte controle van de neurotransmitters in de synaptische terminal doorlive cell imaging technieken fluorescente indicatoren, waaronder exocytose van synaptische blaasjes bekeken styrylkleurstoffen amfipatische FM kleurstoffen (FM1-43 en FM4-64) en / of synapto-pHluorin en specifieke neurotransmitter fluorescente reporters, zoals CNiFERs van ACh en iGluSnFr voor glutamaat 18-20. Kortom, deze de huidige benaderingen voor de identificatie van pre-synaptische lokalisatie van nAChRs zijn ingewikkeld en vereisen speciale systemen en technieken voor betrouwbare identificatie en fysiologische bewaking van pre-synaptische activiteit mogelijk.
Hier beschrijven we protocollen en apparatuur voor een in vitro co-kweeksysteem van een ventrale hippocampus (vHipp) – nucleus accumbens (NACC) schakeling die directe toegang tot het identificeren en zowel pre- en postsynaptische elementen van synaptische transmissie analyse verschaft. We tonen voorbeelden van pre-synaptische lokalisatie van nAChRs en de live-cell imaging van nAChR gemedieerde Ca 2+ signalering en neurotransmitter vrijlating along vHipp axonen. Een natuurlijke (en eenvoudige) uitbreiding van het protocol hier gepresenteerde is de bereiding van pre- en post-synaptische contacten omvat neuronen van verschillende genotypen. Op deze wijze wordt de bijdrage van een bepaald genproduct aan de pre- en / of postsynaptische mechanismen van modulatie kan direct worden bepaald.
De co-kweek bereiding beschreven re-capituleert ventrale hippocampus-accumbens circuits in vitro. Dit preparaat vergunningen betrekkelijk eenvoudige en betrouwbare behandeling van de ruimtelijke en temporele profielen waardoor activatie van presynaptische nAChRs wekken verhoogde glutamaterge transmissie 5, 21.
